Monitoring pilot-scale polyhydroxyalkanoate production from fruit pulp waste using near-infrared spectroscopy

Given the current plastic disposal global crisis, biodegradable polymers may have an important role in leading way towards a more sustainable industry. Polyhydroxyalkanoates (PHA) are biobased and aliphatic polyesters synthesized by bacteria stored as intracellular granules. In this study, PHA was produced at pilot scale mixed microbial culture (MMC) fed with fermented fruit pulp waste. The accumulation reactor monitored line near-infrared (NIR) spectroscopy to develop optimize partial least squares (PLS) calibration models for predicting bulk concentration content. PLS were subjected internal cross-validation (62 spectra) ones best performance validated external test set (12 spectra). For concentration, root mean squared error of prediction (RMSEP) 0.69 g/L coefficient determination (R2) 0.89 attained, while content predicted RMSEP 14.6% R2 0.86. These NIR-based demonstrated great potential real-time monitoring MMC pilot-scale production from complex waste substrate. This approach can be used situ control pulse-wise feeding strategy stage, minimizing consumption triggered carbon source depletion contributing improved process efficiency productivity.
